Venezia Mestre is the principal mainland station serving the Venice metropolitan area, acting as a gateway for passengers who prefer to stay on the mainland or need to connect onward by bus and tram rather than crossing the lagoon. It handles a high volume of regional and intercity traffic, with many long-distance trains terminating here rather than continuing across the causeway to Venezia Santa Lucia. The station serves the sprawling industrial and residential districts of Mestre and provides good connections to Marco Polo Airport via bus.

Regional trains run from Mestre to Venezia Santa Lucia terminus in approximately eight to ten minutes, with multiple departures every hour. Buses also cross the causeway to Piazzale Roma at the entrance to Venice island.
Mestre is a practical and significantly cheaper base than Venice island for accommodation, with fast and frequent train connections to Santa Lucia. Many budget and mid-range hotels are clustered around the station.
Many Frecciarossa and Frecciargento high-speed services that continue to Venice stop at Mestre before crossing the lagoon to Santa Lucia. Euronight and other overnight trains also serve Mestre, as not all of these continue onto the island.
